Vitamin D - Synthesis and Roles in the Body

This is a brief overview of how Vitamin D is synthesised in the body, how it acts on the vitamin D receptor and what effects it has on the body. Vitamin D can be made in the skin when it's exposed to sunlight. First cholesterol is turned into 7 dehydrocholesterol by an enzyme called dhcr7. UVB radiation from the sun, then turns this into previtamin D, which spontaneously transforms into vitamin D at body temperature. This actually means that because Vitamin D can be made by the body, it's technically not a vitamin, since a vitamin by definition is something that can't be made by the body. There are 5 forms of vitamin D found in nature, but the body only makes one, Vitamin D3, also called cholecalciferol. Even when vitamin D has been synthesised from cholesterol, it is still not active, and there are two more steps needed until it becomes active. The first step is the conversion to calcidiol, which happens in the liver. Calcidiol is then converted to calcitriol which is the active form of vitamin D. This step happens primarily in the kidney and is controlled by parathyroid hormone, calcium and phosophate concentrations in the blood, and the concentration of calcitriol in the blood. Now that our vitamin D is finally active it circulates in the blood and acts like a hormone. When it reaches the cell, it binds to the vitamin D receptor also called the calcitriol receptor. This whole thing then forms a big complex with the retinoid X receptor, or RXR receptor. Finally the RXR receptor needs to be bound by Vitamin A, making a big structure composed of calcitriol, the vitamin D receptor, the RXR receptor and vitamin A. This complex won't be active until Vitamin A is present, which effectively means that Vitamin D is useless unless Vitamin A is also available in the cell. When activated, this complex binds specific areas of DNA and increases transcription of some dna and decreases transcription of other dna. functions of vitamin D Probably the most famous role of vitamin D is to increase the level of calcium in the blood. it does this by Promoting absorption of calcium from the gastrointestinal tract, meaning that we get more calcium from the food that we eat Reducing the amount of calcium that is lost in the urine, by Increasing reabsorption in the kidneys stimulating calcium release from bone by activating osteoclast cells Vitamin D also has effects on the parathyroid gland. It acts to stop parathyroid cells from proliferating too much. when the parathyroid gland becomes vitamin D deficient, it starts to grow to a bigger size than it is meant to and secretes too much parathyroid hormone, resulting in hyperparathyroidism. Vitamin D stops parathyroid cells from proliferating too much Vitamin D also has a role in the immune system with vitamin D deficiency and excess both having negative effects. Macrophages, dendritic cells, B cells and T cells can respond to and synthesise calcitriol. Earlier I said that the conversion of calcidiol to calcitriol happens primarily in the kidney but another place that this can happen is in immune cells. Vitamin D can also lead to the production of antibacterial peptides such as cathelicidin and beta defensin 4. These work by disrupting the cell membranes of bacteria, causing them to die. Summary So, in summary, the skin produces vitamin D from cholesterol when exposed to UVB sunlight. Vitamin D is converted to Calcitriol and is then active The most well known role of calcitriol is in calcium metabolism, but it has many other effects, such as in the immune system and on the parathyroid gland Future videos are going to look the roles of vitamin D in more detail, the amount of vitamin D that humans need and how the recommended daily allowance has been determined, the studies that have been done on vitamin D supplementation, what happens in deficiency and excess, and how prevalent vitamin D deficiency is.

I'm gonna break it down super, super simple, super, super fast, not like my normal videos. Okay, number one, more weight training the right way. The thing is, is exercise itself produces testosterone, helps hormones out, and contrary to popular belief, but what a lot of people will tell you, it's not just a matter of weight training only. Cardio itself also boosts testosterone, but when it comes to weight training, we wanna be focusing on heavier compound movements. I'm talking about dead lifts, I'm talking about bench press, I'm talking about squats, simply because it causes a secretion of testosterone later on down the line because of the recovery. Now, here's something kind of interesting. What actually happens is right after your weight training, your testosterone levels plummet. A lot of people will tell you that testosterone levels increase right after you workout, wrong. They decrease for quite some time. There's one study that took a group of participants over the course of four weeks, they had them implement heavyweight training. Well, 40% increase in testosterone after four weeks of introducing heavy compound movements, 40%, that's huge. Additionally, if cardio's your game, do some high intensity interval training. Simply because intermittent fasting works on something called leptin. Leptin is a hormone that is involved in the hunger process of our body, so when we eat, leptin is produced. Leptin also helps us burn some fat. When we down regulate leptin, we can actually allow our bodies to burn more fat, but also boost a little bit of testosterone. Intermittent fasting causes a decrease in body fat, and there's also direct corelation between lower body fat and higher T levels. It constantly exponentially grows from there. Then lastly, is sleep. The big one that I talked about at the very beginning. Studies are now showing that if you are lacking sleep, you could be suffering from a 60% loss of testosterone. Those that got four hours of sleep verses eight hours of sleep, the ones that got four hours had 60% less testosterone than those that got eight. So yeah, you wouldn't think that sleep plays such a big role. Make sure you're getting that sleep, make sure you're getting those fasting days in now and then, make sure that you're focusing on your vitamin D and your sunlight, and also make sure you are lifting heavy. UPDATE 8.1.15 : Unfortunately, I will be repeating testing for my vitamin d, because the test I was given in this vid was not extremely sensitive. It would only show if had an extreme vitamin d deficiency leading to osteoporosis or a parathyroid issue, which says a lot, but not enough. I will be repeating with a more accurate 25 OH vitamin d test. Nevertheless, there is studies on tanning they have high vitamin d levels compared to non-tanners. Glucocorticoids such as Prednisone and Prednisolone can cause osteoporosis within 3-6 months of use and lead to fractures, especially vertebral fractures. Nearly 5% of women in the US are on these drugs for a variety of conditions such as rheumatoid arthritis, asthma, inflammatory bowel disease, vasculitis, and a myriad of skin disorders. The risk is related to the dose. Preventive treatment should include calcium, protein, vitamin D, vitamin K, strontium, regular weight bearing exercise and withdrawl of smoking and alcohol. Low testosterone can lead you to feel fatigued, having a low libido and even have trouble gaining muscle. In a study published in the Journal of Trace Elements in Medicine and Biology in 2011, boron supplementation was given to male volunteers and was paired with steroid hormone level measurements, looking at the impact hourly, daily and weekly. After one week of supplementation: -Free testosterone increased -Estrogen levels decreased -Dihydrotestosterone levels increased -Vitamin D levels increased -Decrease in SHBG six hours after supplementation -Inflammation markers decreased Testosterone is useable in the body when it is free. Most testosterone in the body is bound to the sex hormone binding globulin (SHBG). Through the decrease in SHBG it is likely that testosterone is freed into the blood in the useable form. Vitamin D is also positively correlated with testosterone levels, so by increasing vitamin D in plasma testosterone would increase as well. The same researchers conducted a study in 1997 that found boron supplementation at 10 mg/day in men increased both testosterone and estrogen levels after four weeks. In this study, it was found that only 84% of the boron that was taken was found in the urine. This suggests that the remainder was used in the body. This is a high level of bioavailability. Just as in these studies, the studies that have been conducted have found varying effects on estrogen, some showing it decreasing, some increasing, and some showing no effect. Another study published by BioImmersion Inc. in 2006 found that men who supplemented with boron in the form of fructo borate for two weeks experienced increased levels of vitamin D and testosterone levels. -Free testosterone increase of 29.5% -Vitamin D3 increase of 19.6% -Estrogen changes varied with no established trend -DHEA increase of 56% Tips: Minerals are not easily passed out of your body, so you want to be careful not to take too much. You do not want to take too much boron as it may become toxic at greater than 20 mg per day, which is set as the Tolerable Upper Limit (TUL) of boron. One study on male bodybuilders found no increase in testosterone levels when supplementing for 7 weeks with 2.5 mg of boron, so it seems that higher than this may be necessary to see results. Recommended daily supplementation ranges from about 6 mg to 20 mg per day. You can also increase your dietary intake of boron through consuming organic fruits and veggies, including fruits, vegetables, legumes and tubers. References: 1.
